As a novel minimally invasive approach, microneedle-mediated transdermal drug delivery has received increased attention. Microneedle patch effectively penetrates the skin barrier of the stratum corneum and create efficient pathways for the delivery of drugs.

A randomized controlled trial based on split-body was planned to evaluate the efficacy of enhanced transdermal delivery of topical agent by hyaluronic acid -fabricated microneedle patch in psoriatic plaques. Six psoriatic plaques in a patient will be randomized to either microneedle HA patch, patch, or control groups. Patch group is planned for rule out the occlusion effect of patches. All lesions will be treated with topical agent and patches daily for 2-week period. The severity of psoriasis will be grated with modified PASI score and the clinical improvement will be assessed as % from baseline every week. Overall 20 patients with 120 psoriatic plaques will be enrolled.

Effect Of The Tested Formulations On Hacat Cell Apoptosis

Cells were seeded in 6-well plates at 2 × 105 per well, cultured for 24 h, and were either not treated further or treated with TNF- at a final concentration of 100 ng/mL. An equal volume of PBS was added to the control group. After incubation for 2 h, the tested formulations were treated with curcumin at a final concentration of 8.0 µg/mL, and incubation was continued for 46 h. Cells were incubated with 0.25% trypsin for 5 min, washed with serum-free medium, washed with PBS, and suspended with binding buffer. Blinded observers reported the benefits in 40 women who applied topical cross-linked hyaluronic acid twice a day to their full face, compared with 20 women who applied the nanotechnology vehicle only. Additional significant improvements were seen among another 40 women who applied non-cross linked topical hyaluronic acid using the same regimen.

“We know the skin is a barrier. It acts as a barrier for things getting in, but it can also prevent topical compounds from getting in,” said Dr. Glynis R. Ablon, who is on the dermatology faculty at the University of California, Los Angeles.

“Topical hyaluronic acid does appear to be penetrating the skin with this unique Ionic Nano Particle Technology delivery system,” she said.

Dr. Ablon and her colleague Dr. Mark S. Nestor conducted the study at two U.S. sites. Participants were 35 to 65 years old and evaluated at days 15, 45, 90, and 120 .

Male C57BL/6 mice were provided by the Experimental Animal Center of Shanghai University of Traditional Chinese Medicine, and the protocol used was approved by the Experimental Animal Ethics Committee of Shanghai University of Traditional Chinese Medicine .
